Role of mitochondrial YAP and TAZ in muscle growth, regeneration and mechanotransduction
proposé par Mario PENDE, INEM, InsermU1151
Projet de stage :
YAP and TAZ are transcriptional co-regulators sensing cell contractility and
promoting tissue growth through the expression of nuclear target genes involved in
cell cycle and survival. We have preliminary evidence that YAP and TAZ are also
localized in the mitochondria. The student will use imaging, metabolomics and
biochemical purification to analyze the role and regulation of mitochondrial YAP/TAZ.
These studies may suggest a powerful mechanism in the cell to coordinate metabolic
responses in nucleus and mitochondria. Mechanical cues could therefore be
transduced by mitochondrial YAP/TAZ in order to stimulate ATP production for cell
contractility.
Techniques mises en œuvre par le stagiaire : confocal immunofluorescence, metabolomic analysis,
purification of mitochondria, gene expression
